Exemestane is a 17-oxo steroid that is androsta-1,4-diene-3,17-dione in which the hydrogens at position 6 are replaced by a double bond to a methylene group. A selective inhibitor of the aromatase (oestrogen synthase) system, it is used in the treatment of advanced breast cancer. It has a role as an antineoplastic agent, a xenobiotic, an EC 1.14.14.14 (aromatase) inhibitor and an environmental contaminant. It is a 17-oxo steroid and a 3-oxo-Delta(1),Delta(4)-steroid. It derives from a hydride of an androstane.

/LABORATORY ANIMALS: Chronic Exposure or Carcinogenicity/ A 2-year carcinogenicity study in mice at doses of 50, 150 and 450 mg/kg/day exemestane (gavage), resulted in an increased incidence of hepatocellular adenomas and/or carcinomas in both genders at the high dose level. Plasma AUCs (0-24 hr) at the high dose were 2575 +/- 386 and 5667 +/- 1833 ng·hr/mL in males and females.
