T-cell surface glycoprotein CD3 gamma chain, also known as CD3G, is a 20 kDa single-pass type I membrane protein that is part of the TCR-CD3 complex. CD3 gamma forms a heterodimer with CD3 epsilon which associates with TCR beta, while CD3δ/CD3 epsilon heterodimer associates with TCR alpha. CD3 gamma /CD3 epsilon /TCR beta joins CD3δ/CD3 epsilon /TCR alpha and recruits a CD3 zeta homodimer to form the complete TCR-CD3 complex. CD3 chains contain one cytoplasmic ITAM domain which mediates intracellular signal transduction. The extracellular domain of CD3 gamma contains one disulfide bond and 2 N-linked glycosylation sites. The ECD of human CD3 gamma shares 60% and 62% sequence identity with mouse and rat CD3 gamma, respectively. The biologic roles of each CD3 chain and their relative contribution to the signals transduced through the TCR/CD3 complex and to downstream activation events are still uncertain, whether they may be specialized or redundant. Deficiencies in CD3 gamma can cause impairments in immune function, particularly in Treg cells which may contribute to autoimmune disorders. Post-translational: Phosphorylated on Tyr residues after T-cell receptor triggering by LCK in association with CD4/CD8 (PubMed:2470098). Phosphorylated also by PKC; leading to the TCR complex down-regulation (PubMed:8187769).Phosphorylated on Tyr residues after T-cell receptor triggering by LCK in association with CD4/CD8.
